1．操作步骤
（1）细胞染色前，准备3种贮存液：①NBT：用10ml 70％DMF溶解0．5gNBT；②BCIP：用10ml 100％DMF溶解0．5g BCIP[二钠盐儿③碱性磷酸酶缓冲液：100mmol／LNaCl，5mmol／LMgCl2，100mmol／LTris（pH 9．5）。所有的贮存液置4C可保存1年。
（2）实验前，底物液新鲜配制。加66／ul NBT贮存液至10ml碱性磷酸酶缓冲液中，充分混合，加33ul BCIP贮存液，在1h内使用。
（3）将洗涤过的标本片放在一个适当的容器内。加足够量的底物液覆盖标本片（3～5ml适合于100mm平皿）。在室温中进行反应直到染色达到适当黑色。对一些试剂可在显微镜下定期监测。一般孵育时间大约是30min。
（4）在含有20mmol／LEDTA的PBS中漂洗以终止反应，螯合Mg2+。
（5）优化：如果需要，进行复染。
（6）用水冲洗，DPX封片。
2．需要的溶液和特殊设备
碱性磷酸酶缓冲液：100retool／L NaCl，5mmol／L MgCl2，100mmol／L Tris（pH9．5）；
NBT溶液（0．5gNBT用10ml 70％DMF溶解，贮存在4C）；
BCIP溶液（0．5gBCIP[二钠盐]用10m1100％DMF溶解，贮存在413）；
20mmol／LEDTA用PBS溶解，DPX。
