1、 制备含MMP底物蛋白的SDS-PAGE凝胶：推荐分离胶浓度为8%。按照标准程序制备SDS PAGE凝胶。将10 × substrate G 融化，并90 ºC 加热5分钟。分别在浓缩胶和分离胶中额外加入10 × substrate G 并使之稀释10倍，混匀后加入过硫酸铵和TEMED，等待凝胶聚合。
2、待测样品1:1 稀释于2 × SDS-PAGE non-reducing buffer (用完后可自行配制：4% SDS, 100 mM Tris-Cl pH6.8, 20% glycerol, 0.02% bromophnol blue)，混合后直接上样。切勿加热变性，并且仅使用不加还原剂的上样缓冲液。可通过预试验确定加样量，使用普通蛋白预染Marker 即可。
