用标记了FITC的AnnexinⅤ作为荧光探针，利用流式细胞仪或荧光显微镜可检测细胞凋亡的发生。细胞坏死的过程中也会发生细胞膜损伤，坏死的细胞会结合Annexin V-FITC。正常细胞和早期凋亡细胞的细胞膜是完整的，Propidium iodide（PI）是一种核酸染料，它不能透过完整的细胞膜，但在凋亡中晚期的细胞和坏死细胞，PI能够透过细胞膜与细胞核结合呈现红色。将AnnexinⅤ与PI匹配使用，可以将凋亡早期的细胞和晚期的细胞以及死细胞区分开来。坏死细胞可以同时与Annexin V-FITC 和PI 结合显色，而PI 则被排除在活细胞（FITC 阴性）和早期凋亡细胞（FITC 阳性）之外。在没有巨噬细胞的情况下，凋亡的后阶段会像细胞坏死一样发生整个细胞的解体，从而使凋亡晚期的细胞也同时被FITC 和PI 结合染色呈现双阳性。

在双变量流式细胞仪的散点图上，左下象限显示活细胞，为（FITC-/PI-）；右上象限是非活细胞，即坏死细胞，为（FITC+ /PI+ ）；而右下象限为凋亡细胞，显现（FITC+ /PI-）。（图1)
